经胎盘接种小鼠巨细胞病毒宫内感染模型建立 被引量:9

Primary studies on the establishment of murine cytomegalovirus intrauterine model.

摘要 目的应用小鼠巨细胞病毒宫内感染模型,观察母鼠妊娠结局。方法用ELISA筛选无MCMV感染史性成熟期小鼠,雌雄同笼受孕。孕龄12.5天时,模型组胎盘显微注射MCMV,对照组注射生理盐水,空白对照组不做任何处理。部分在孕龄18.5天处死母鼠,部分待母鼠自然分娩,观察母鼠妊娠结局及子代感染情况。结果模型组36.62%(26/71)和生理盐水对照组29.85%(20/67)的死胎率,χ2=0.711,P>0.05。分别和空白对照组0.00%(0/68),P<0.01。比较三组组畸胎率分别为22.54%(16/71)、0.00%(0/67)和0.00%(0/68),P<0.01。空白对照组和生理盐水对照组未发现胎盘和胎仔感染,模型组胎盘感染率63.27%(31/49)、胎仔感染率为32.65%(16/49),χ2=0.136,P<0.01。结论小鼠巨细胞病毒宫内模型的建立为其发病机理研究、选择宫内感染的干预介入点提供了新的靶点。 Objective: Study the pregnangcy outcome after intrauterine murine cytomegalovirus ( MCMV ) infection. Methods: The mature mice without MCMV infection identified by Enzyme -linked immunoabsorbent assay (ELISA) were served as the subjects. We put the male and the female in the same cages, and then we injected MCMV into the placentas of the model group mice at day 12. 5 of gestation, the saline control group were injected with saline, the blank control group were not do any treatment. The conditions of the development and infections of embroys were allowed to observe until day 18. 5 of gestation. Results: The still- born rate of the model group and the saline group were 36. 62% (26/71 ) and 29. 85% (20/67), x^2 = 0. 711, P 〉 0. 05. but The still - born rate of the blank group were 0. 00% , P 〈0. 01. neither infection placenta nor embryonic infection was found in the saline or blank control group, while the rate of placenta infection in the model group the were 63. 27% (31/49) and the embryonic infection 32. 65% ( 16/49), P 〈 0. 01. The still- born rate of two groups were 31.11% (14/45) and 34. 69% (17/49), x^2 = 0. 136, P 〉 0. 05. respectively, and significant difference was found among the three groups. Conclusion: The establishment of murine cytomegalovirus intrauterine model may make for the study of pathogenesis and treatment.
