摘要
目的:研究不同强度运动训练对大鼠海马CA1区神经元凋亡的影响及其基因调控机制。方法:将大鼠分为对照组、中等强度运动组和大强度运动组,对大鼠进行为期8周的游泳训练,用DNA原位末端标记法检测海马神经元的凋亡,并用免疫组化的方法观察海马神经元中bcl-2、bax的免疫反应活性。结果:(1)大强度运动训练后,大鼠海马CA1区神经元凋亡显著增加而中等强度运动训练后,大鼠海马CA1区神经元凋亡不明显,海马神经元凋亡可能是大强度运动训练导致运动能力降低和中枢性运动疲劳的病理生理机制;(2)大强度运动训练后,大鼠海马CA1区神经元中bcl-2蛋白的表达显著下降,bax蛋白的表达显著增加。中等强度运动训练后,大鼠海马CA1区神经元bcl-2蛋白的表达显著上升。因此,大强度运动训练可抑制海马神经元bcl-2蛋白的表达而促进bax蛋白的表达,这可能是大强度运动训练导致大鼠海马CA1区神经元凋亡发生的基因调控机制,而中等强度运动训练可促使bcl-2蛋白的表达上升,抑制细胞凋亡。
Objective:Examine the influence of different intensity sport training on neuron perishing of the big mice hippocampus CAI area and its gene regulation mechanism. Method: The mice were divided into the control group, the medium intensity sport group and the great intensity sport group, carries on to the big mouse for 8 week-long swimming training, weakly perishes with the DNA home position terminal sign notation examination hippocampus neuron, and observed in tbe hippocampus neuron with the immunity group's method bcl-2, the bax immune response activeness. Results: After the great intensity movement training, and after the big mouse hippocampus CA1 area neuron weakly perishes the significant increase but under the medium intensity training, the perishing is not obvious.The hippocampus neuron weakly perishes possibly is the great intensity movement training causes the movement ability to re- duce with the main center movement weary pathology physiology mechanism. After great intensity movement training, the bcl-2 protein expression remarkable drop, but bax protein expression remarkable increase. After medium intensity movement training, the bcl-2 protein expression remarkable rise. Therefore, the great intensity movement training may suppress the hippocampus neuron bcl-2 protein the expression but to promote the bax protein expression, this possibly is the great intensity movement training causes the gene regulation mechanism which the big mouse hippocampus CA1 area neuron weakly perishes occurs, but the medium intensity movement training may urge the bcl-2 protein the expression rise, the suppression cell weakly to perish.
