肿瘤-睾丸抗原SSX_1和SSX_4 mRNA在肝细胞癌中的表达

摘要 目的检测肿瘤-睾丸抗原(CTA)SSX1和SSX4mRNA在肝细胞癌(HCC)及外周血中的表达,探讨CTA抗原SSX1和SSX4在HCC中表达有无特异性。方法应用逆转录-聚合酶链反应(RT-PCR)方法对35例HCC患者癌组织和相应的癌旁组织SSX1和SSX4基因表达进行检测,对其中6例RT-PCR扩增产物的目的片段进行DNA序列测定;HCC确诊后第2周对30例患者外周血中SSX1基因进行检测。结果35例HCC患者中,SSX1和SSX4的表达率分别为77.1%(27/35)和65.7%(23/35);癌旁组织、12例肝硬化和15例正常肝组织未检测到SSX1和SSX4的表达。6例DNA序列测定结果显示RT-PCR产物为SSX1和SSX4的cDNA。SSX1和SSX4的表达与患者年龄、性别、肿瘤大小、血清甲胎蛋白(AFP)水平、乙型肝炎病毒(HBV)和丙型肝炎病毒(HCV)感染无相关关系(均P>0.05)。外周血中未检测到SSX1表达。结论SSX1和SSX4在HCC中呈高度特异性表达,是制备HCC疫苗理想的靶向分子,多种CTA联合表达为多效价疫苗治疗HCC提供了理论基础。 Objective To investigate the mRNA expression of SSX1 and SSX4 in human hepatocellular carcinomas (HCCs) and peripheral blood, and to study their specificity in HCCs. Methods Rcverse-transcription polymerase chain reaction (RTPCR) was used to detect the expression of the SSX1 and SSX4 genes in cancerous and corresponding para-cancerous liver tissues of 35 cases of HCC. Among them. 6 cases were subjected to the DNA sequencing of the amplification products. On the second week after diagnosis of HCC the expression of SSXr gene in peripheral blood was detected in 30 cases of HCC simultaneously. Results In the 35 cases of HCC, the expression rate of SSX1 and SSX4,mRNA was 77.1% and 65.7 % respectively. In corresponding para-cancerous tissues. 12 hepatocirrhosis tissues and 15 normal liver tissues, the expression of S SSX1 and SSX4 was undetectable. DNA sequencing showed that the RT- PCR amplification products were SSX1 NA in 6 cases. The expression of SSX1. rgenes was not correlated with age. sex. tumor size. serum AFP. and HBV and HCV infection (P〉0.05). In peripheral blood no SSX1 was detectable. Conclusion SSX1 mRNA expression was highly specific in HCCs which would not only provide the ideal target molecular sites for HCC tumor vaccines but also establish the potential value of the polyvalent tumorantigen vaccines for HCC therapy.
