摘要
目的研究哮喘大鼠肺组织TRAIL的表达及支气管肺泡灌洗液(BALF)中T淋巴细胞凋亡情况,以及氨茶碱对其的影响。方法将大鼠分为4组:正常对照组、哮喘组、氨茶碱组及地塞米松组。应用免疫组织化学结合图像定量分析方法对TRAIL蛋白表达进行检测。急性分离大鼠BALF中T淋巴细胞,在植物凝聚素-P(PHA-P)的刺激下培养2 h,经流式细胞仪检测大鼠BALF中T淋巴细胞凋亡率。结果大鼠肺组织TRAIL呈组成性表达,哮喘时,其表达明显增加(P<0.05),氨茶碱可以减少哮喘大鼠肺组织TRAIL表达(P<0.05)。哮喘大鼠BALF中的T淋巴细胞凋亡率较正常大鼠明显下降(P<0.05),氨茶碱可以促进哮喘大鼠BALF中的T淋巴细胞凋亡(P<0.05)。结论氨茶碱可能通过抑制肺组织TRAIL的表达来抑制TRAIL的促炎作用,同时还可以通过促进哮喘大鼠气道的T淋巴细胞凋亡发挥抗炎作用。
Objective To explore the expression of TFRAIL in asthmatic rat lung and the apoptosis of T-lymphocytes in asthmatic rat BALF and the effect of aminophyline on them. Methods The rats were divided into normal control group, asthmatic group, aminophyline-treated group and dexamethason-treated group. Immunohistochemistry in combination with imaging quantitative analysis method was used for detection of the expression of TRAIl. protein. The isolated T-lymphocytes from BAI.F of rats were cultured for 2 h under the stimulation of PHA-P. The apoptosis of the T-lymphocytes was assayed by flow eytometry. Results TFRAIL was expressed constitutively in normal rat lung. The expression of TRAIl. was increased greatly in asthmatic rat lung (P〈0.05) and aminophyline could decrease the expression of TRAIL in asthmatic rat lung (P〈0.05). The apoptotic rate of T-lymphocytes in asthmatic rat BALF was significantly lower than that in normal rat BALF, and aminophyline could promote the apoptosis of T-lymphocycs in asthmatic rat BALF Conclusion Aminophyline could inhibit the inflammation of asthmatic airway by reducing the expression of TRAIl. in rat lung and promoting the apoptosis of T-lymphocytes in asthmatic rat airway.
