血管内皮生长因子及碱性成纤维细胞生长因子在喉癌细胞Hep-2中的表达 被引量:1

摘要 目的研究血管内皮生长因子(vascular endothelial growth factor,VEGF)和碱性成纤维细胞生长因子(basicfibroblast growth factor,bFGF)在喉癌细胞Hep-2中的表达,及其与喉癌细胞生长、浸润及转移的关系。方法培养Hep-2和正常永生化表皮细胞HaCaT,提取总细胞RNA,应用逆转录聚合酶链反应检测VEGF mRNA和bFGF mRNA在Hep-2和HaCaT细胞中的表达。结果VEGF mRNA在Hep-2细胞中高表达,平均相对积分吸光度值为(0.839±0.063);VEGF mRNA在HaCaT细胞中低表达,平均相对积分吸光度值为(0.305±0.066),差异有极显著性意义(t=14.94,P<0.01)。bFGF mRNA在Hep-2细胞中高表达,平均相对积分吸光度值为(0.792±0.058);bFGF mRNA在HaCaT细胞中低表达,平均相对积分吸光度值为(0.296±0.049),差异有极显著性意义(t=17.54,P<0.01)。结论VEGF及bFGF与喉癌细胞的生长、浸润及转移有密切关系。 Objective To study the expression of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) in human laryngeal carcinoma cells (Hep-2) and its relation with growth, invasion and metastasis of laryngeal cancer cells. Methods The total RNA was isolated from the cultured Hep-2 and human normal keratinocyte line HaCaT. The mRNA expression of VEGF and bFGF in Hep-2 and HaCat cells was detected by using technique of reverse transcriptase polymerase chain reaction. Results Image analysis demonstrated that the expression levels of VEGF mRNA in Hep-2 cells were higher than in HaCaT cells with the corrected integral absorbency being (0.839±0.063) and (0.305±0.066) respectively. There was significant difference between them (t= 14. 94. P〈0. 01 ). The expression levels of bFGF mRAN in Hep-2 cells were higher than in HaCaT cells with the corrected integral absorbency being (0. 792±0. 058) and (0. 296±0. 049) respectively. There was significant difference between them (t=17. 54. P〈0.01 ). Conclusion VEGF and bFGF was closely related with the growth, invasion and metastasis of laryngeal carcinoma cells.
