摘要 目的探讨Clara细胞分泌蛋白10-kDa(CC10)在苦参碱诱导的肺腺癌细胞凋亡中的表达。方法肺腺癌A549细胞进行传代培养72 h后,分别加入30、60、120、2404、80 mg/L的苦参碱,MTT法检测其对A549细胞生长的抑制作用,流式细胞仪检测细胞凋亡,Western blot及RT-PCR检测CC10蛋白及mRNA的表达。结果不同浓度的苦参碱对肺腺癌细胞生长均有抑制作用,并呈浓度依赖性。实验组细胞凋亡率和CC10基因表达率高于对照组,也呈浓度依赖性。结论CC10基因表达增高可能在苦参碱诱导肺腺癌A549细胞凋亡中起重要作用。 Objective To study the expression of Clara cell secretory protcin 10-kDa (CC10) in the matrinc-induced apoptosis of lung adenocarcinoma. Methods After lung adenoearcinoma A549 cells were cuhured for 72 h. matrine of 30. 60. 120, 240 and 480 mg/L was added separately, The inhibitory effects of matrine on the growth of A549 cells were tested by using MTT method, apoptosis was detected hy flow cytometry, and the expression of CC10 protcin and mRNA by Western blot and RT-PCR, respcctivcly. Results Matrinc with diffcrent concentrations had inhihitory effects on the growth of lung adenoeareinoma cells in a concentration dependent manner. In the experimental group, apoptotic ratc and the positive expression rate of CC10 gene were. higher than in the control group in a conccntration dependent manner also. Conclusion The overexpression of CC10 might play an important role in the matrine induced apoptosis of lung adenocarcinoma A519 cells.
