在心房颤动心房中Calpain-I与细胞凋亡的关系和意义 被引量:1

The relationship between calpain-I and apoptosis at the left fibrillating human atria

摘要 目的探讨CalpainI蛋白和细胞调亡在心房结构重构中的关系和作用。方法选择行换瓣手术的风湿性心脏病患者,在外科手术中取其左心房组织,用免疫印迹法测定calpainI蛋白含量;用TUNEL法检测左心房肌细胞凋亡,计算其凋亡指数(AI)。分析比较它们之间的关系。结果在慢性心房颤动(房颤)患者中,calpainI蛋白含量明显增加(344%±101.9%,P<0.001);AI明显升高(24.6%±9.1%,P<0.01)。CalpainI的蛋白含量和AI分别与左心房内径、房颤持续时间呈明显正相关(P<0.05~0.001);且CalapinI的蛋白含量与AI亦呈明显正相关(P<0.001)。结论慢性房颤时左心房组织中的calpainI蛋白含量和心肌细胞凋亡均增加,两者之间有协同作用,恶化心房的结构和功能,促进房颤的发生。 Objectives To investigate the relationship between the proteinase calpain-Ⅰ and apoptosis and the significance of these factors for the atrial structural remolding. Methods The left atrial tissues of patients with rheumatic heart disease ( RHD), who required valve-replacement, were collected in surgical operating. The protein content of calpain-Ⅰ was examined by Western blotting. The apoptosis index (AI)were measured through TUNEL. The relationship between the two factors was analyzed. Results In patients with chronic atrial fibrillation(CAF) ,the protein content of calpain-Ⅰ and the AI all increased to 344% ± 101.9% and 24.6% ± 9.1% respectively ( P 〈 0.01 ; P 〈 0.001 ). The protein content of calpain- I and apoptosis index (AI)were positively relative to left atrial dimension and atrial fibrillation duration, respectively( all P value 〈 0.05 ) ; (4) The protein content of calpain-Ⅰ positively related with that of AI ( P 〈 0.001 ). Conclusions In chronically fibrillating left atira, apoptosis of atrial cell and the protein content of calpain-Ⅰ increased. They had cooperative action to deteriorate the atrial structure and dysfunction for promoting the onset and maintenance of atrial fibrillation.
