摘要
目的观察罗格列酮(R os ig litazone,RO S)对肾小管上皮细胞整合素水平的影响。方法将体外培养的大鼠近端肾小管上皮细胞分为对照组、正常血糖组(N)、高糖组(H)、正常培养液+罗格列酮组(RO S)、高糖+罗格列酮低浓度组和高糖+罗格列酮高浓度组六组,采用W estern B lot及RT-PCR了解不同浓度的罗格列酮(5μm o l/L、10μm o l/L)对高糖刺激(30mm o l/L)下整合素1β(In tegrin-beta1)蛋白质水平表达的影响。结果高糖刺激下,In tegrin1β(84710.79±1810.4)较对照组(23283.17±827.55)及正常葡萄糖组(47896.31±2014.63)明显增加(P<0.05),RO S组In tegrin1β(22609.49±1076.26)较高糖组降低,RO S 10μm o l/L组(38603.43±1409.30)的降低程度较RO S 5μm o l/L组(31327.16±1721.36)更明显。结论罗格列酮可明显抑制高糖刺激下肾小管上皮细胞In tegrin1β的表达,且有明显剂量依赖关系。
ObjectiveTo evaluate the effect of rosiglitazone on the level of integrin β1 in proximal tubular cells. Methods The proximal tubular cells of rats cultured in vitro were divided into 6 groups : control group, normal glucose group, high glucose group, high glucose with rosiglitazone of 5μmol/L group, high glucose with rosiglitazone of 10μmol/L group and single rosiglitazone of 10μmol/L group. The cells werecuhured for 48 b. Integrin β1 was detected by Western blot. Results The expression of integrin 61 was significantly increased in high glucose group (84710. 79± 1810. 44)as compared with control group (23283.17±827.55)and normal glucose group(47896.31±2014.63), Integrin β1 in high (30 mmol) dose of glucose group with 5 or 10 mmol roslglitazone was 31327.16±1721.36 and 38603.43± 1409.30, respectlvely(both P〈0. 005). Conclusion Rosiglitazone can strikingly inhibit the expression of integrin β1 in proximal tubular cells exposed to high glucose with a barked dose-dependent manner.
