小鼠胰岛细胞MHC-DR抗原的诱导表达

摘要 本文报告肿瘤坏死因子(TNF)和r-干扰素(IFN-r)诱导小鼠胰岛细胞MHC-DR抗原表达对胰岛素分泌的影响。实验结果表明:(1)当TNF浓度为50~100U/ml时未见胰岛细胞表达MHC-DR抗原,而50U/mlr-IFN可诱导少量胰岛细胞(8.5±2.9%)表达MHC-DR抗原;(2)100U/mlTNF和100U/mlr-IFN合并使用,MHC-DR抗原阳性细胞明显增多(80.25±4.11%);(3)两种因子单独或合并作用均能使体外培养的胰岛细胞的胰岛素分泌量增加(P<0.01)。上述结果提示,胰岛细胞MHC-DR抗原的诱导表达本身并不影响其分泌胰岛素的功能。 The correlation of induced-expression of MHC-DR molecules with insulin release i cul-tured mice islet cells has been studied.The results showed that:(1)No detectable induced-expression of MHC-DR molecules by TNF-a alone at the concentration of 50一100U/ml hasbeen assayed,but the induced-expression of MHC-DR molecules has been identified in a fewislet cells(8.5±2.9%)with IFN-r alone at concentration of-100U/ml.(2)The in-duced-expression of MHC-DR molecules has been detected in as high as 80% of cultured isletcells with TNF-a and IFN-r combined at different concentrations.(3)Insulin released fromcultured islet cells was enhanced by IFN-r and TNF-a either alone or combined at different concentrations.These results suggest that the induced-expression of MHC-DR molecules themselves does not damage the function for insulin release in cultured islet cells.
