摘要
本实验通过给Wistar成年雄性大鼠灌服不同剂量的雷公藤多甙(GTW),检测大鼠睾丸内支持细胞(SC)雄激素结合蛋白(ABP)、硫酸化糖蛋白-2(SGP-2)和间质细胞膜上黄体生成素受体(LH/CG-R)在转录水平上的变化,进一步探讨GTW的雄性抗生育机制。雄性大鼠服GTW剂量分为每日30mg/kg共35d(组I),与服GTW每日10mg/kg共49d(组Ⅱ)。结果表明:两实验组中SC分泌蛋白ABP与SGP-2mRNA水平与对照组相比均有升高,并与喂服GTW剂量呈正相关,ABP的两种mRNA1.7kb与2.3kb)中1.7kbmRNA在实验组中呈升高趋势,而2.3kbmRNA基本无变化。分析间质细胞的LH/CG-RmRNA变化发现,其1.2kbmRNA在实验组中有升高,而4.4mRNA变化不大。同时,对上述变化的机理进行了讨论。
Our studies were undertaken to assess the male antifertility mechanism of the total gly-cosides extracted from Tripterigium Wilbrdii Hook.f.(GTW)./In experimental group I,GTW vvas given by gastric gavage at a dose of 30mg/kg per day for 35 days.In experimentalgroup Ⅱ, GTW was given in the same way at a dose of 10mg/kg per day for 49 days.The transcripts level analysis indicated that in all the experimental groups ABP,SGP-2and LH/CG-R mRNA were increased.Two ABP and two LH/CG-RmRNA transcripts withrespective molecular sizes of 1.7kb,2.3kb and1.2kb,4.4kb were identified.ABp and LH/CG-R mRNA with 1.7k band 1.2kb increased,While those with 2.3 and 4.4kb remained un-changed.In addition,the changes of different proteins at the mRNA level were shown to beGTW dose-dependent.The morphological features of germ cell degeneration and appearanceof germ cells in tubular lumen suggest that the increases in transcripts level of three proteinsin testis may not be directly associated with the effect of GTW,The question whether GTWinterfere with the function of sertoli cells and leydig cells would be study.
