摘要
选用雄性大鼠1次肌注康宁克通-A,小剂量2.1 mg/kg,大剂量10.5 mg/kg。另设阳性对照每日20 mg/kg 氢化泼尼松,肌注4周,并设空白对照。用药后4天及4周末电镜观察胸腺及肾上腺皮质。小剂量组无明显变化,大剂量组及阳性对照组均见到胸腺淋巴细胞数量明显减少、胞核固缩、细胞崩解,上皮性网状细胞及巨噬细胞增多。肾上腺皮质束状带变薄,细胞线粒体及脂滴减少。提示,大剂量用药对肾上腺皮质和胸腺等有损伤作用,临床用药在达到疗效前提下,尽量减少用药量为好。
Two groups of young male rats were investigated in this experiment.The 2 groups of rats were injected with 2.1 mg/kg and 10.5 mg/kg Kenacort-A respectively.Besides the 2 groups of rats,a positive contral group which were injected with 20 mg/kg hydroprednisone and a negative con- tral group were observed.After 4 days and 4 weeks the thymus and adrenal glands were studies by electron microscope.The results showed that there were no remarkable changes in the rats injected with low dosage kenacort-A.In the high dosage group and positive control group the number of thymic lymphocytes was reduced markedly.Pyknoitic nuclei and lysis of thymic lymphocytes were observed. The number of thymic epithelial ceils and macrophages increased in the thymus.The zona fasciculata of adrenal cortex become thin and the number of mitochondria and lipoid granules were reduced in the cell of the zona fasciculata.The results suggest that a high dosage of Kenacort-A could damage the a- drenal cortex and thymus.While the satisfying curative effects were taken in clinic.The dosage of ke- nacort-A should be as little as possible.
