从生物组织中提取 DNA,一般都是选用肝组织,但本文选用小鼠的脾脏,通过粉碎组织、裂解细胞膜与核膜,使核蛋白释放。然后用酚抽提,再经7.5 mol/m^3乙酸钠-异丙醇混合液沉淀,得到白色絮状 DNA,通过O.D_(260)nm/O.D_(280)nm 的比值以及琼脂糖凝胶电泳证实 DNA 是纯的。紫外分光光度计可测定 DNA 含量。

In this paper we report extraction of DNA from mouse spleen.The spleentissue was crushed down.Then the cell membranes and nuclear envelopes were splitted.Nuclear proteins were re- leased.Undigested proteins were extracted with phenol.DNA were the precipitated with 7.5 M sodi- um a cetate:isopropanol mixture was white fluffy mass.DNA was pure as judged by the value of OD_(260 nm)/OD_(280 nm)and agarose gelelectrophoresis.DNA content was determined by ultraviolet spec- trophotometer.
