当归补血汤含药血清对培养内皮细胞生长因子受体表达的影响 被引量:24

摘要 目的探讨当归补血汤含药血清对人脐静脉内皮细胞血管内皮生长因子(VEGF)受体表达的影响。方法将培养的人脐静脉内皮细胞(HUVEC)随机分为6组:①正常对照组;②100μmol/LH2O2低氧培养组;③bFGF+100μmol/LH2O2组;④黄芪、当归液大剂量组+100μmol/LH2O2;⑤黄芪、当归液中剂量组+100μmol/LH2O2;⑥黄芪、当归液小剂量组+100μmol/LH2O2。HUVECVEGF受体的检测采用免疫荧光化学染色法,同时用RT-PCR方法测定VEGFmRNA的表达。结果采用H2O2可成功制备内皮细胞缺氧模型,与正常对照组比较,当归补血汤含药血清各组VEGF受体均有不同程度的表达,其中中剂量组和小剂量组具有明显的上调受体的作用。对于VEGFmRNA的表达,以H2O2低氧培养组、中剂量+H2O2组和小剂量+H2O2组较为显著。结论化学性H2O2损伤可以成功造成HUVEC缺氧损伤,益气活血中药当归补血汤可以上调VEGF受体,以及VEGFmRNA的表达,促进血管新生。 Objective To study effect of serum containing Dangguibuxue Decoction on expression on VEGF receptor in cultured endothelial cell. Methods The cultured HUVEC were divided into 6 groups: Control; 100 μmol/L H2O2 hypoxia group; bFGF + 100 μmol/L H2O2 group; large dose + 100 μmol/L H2O2 group; middle dose + 100 μmol/L H2O2 group; small dose + 100 μmol/L H2O2 group. Immunofluorescence was used to detect HUVEC VEGF receptor, and RT PCR was used to evaluate VEGFmRNA expression. Resuits H2O2 could successfully produce hypoxia. Contrasting to control group,all dose groups could improve expression of VEGF receptors. Middle dose and small dose had a markedly effect of up regulating receptor. With expression of VEGF mRNA,H2O2 hypoxia group, middle dose + 100 μmol/L H2O2 group and small dose + 100 μmol/L H2O2 group improved significantly. Conclusion Chemical injury of H2O2 could make a good hypoxia model of HUVEC. The Dangguibuxue Decoction of nourishing qi and activating blood could up- regulate VEGF receptor, promote the expression of VEGFmRNA, and improve angiogenesis.
