邻苯二甲醛柱前衍生法测定海洋胶体中的水解氨基酸 被引量:8

Determination of Total Hdrolysable Amino Acids with High-performance Liquid Chromatography and Pre-column Derivation with o-phthaldialdehyde in Marine Colloids

摘要 用邻苯二甲醛(OPA)柱前衍生反相高效液相色谱法测定了厦门湾南部海域海洋胶体中氨基酸的组成和含量.该法的空白值为0~1.8×10^-3μmol/L,检出限为2×10^-3~9×10μmol/L.添加脲的回收率在99.5%~99.7%之间,相对偏差为3.4%~10.7%.结果表明,该海洋胶体中含有16种氨基酸。总量为150.83μmol/L,其中赖氨酸是含量最多的氨基酸,浓度为35-59μmol/L,占总水解氨基酸的23.6%.其次是天冬氨酸、谷氨酸、组氨酸、甘氨酸+苏氨酸、甲硫氨酸和亮氟酸,它们的含量相当,浓度分别为12.40、10.48、17.77、16.60、17.22和15.88μmol/L.分别占总水解氨基酸的8.2%、7.0%、11.8%、11.0%、11.4%和10.5%.这8种氨基酸占了胶体中总水解氨基酸的83.5%.其余8种氨基酸只占16.5%. Total hydrolysablc amino acids (THAA) ,the composition and concentration of individual amino acid were measured in marine colloids of Xiamen Bay. For analysis of amino acids,the high-performance liquid chromatography (HPLC) method using precolumn derivatisation with o-phthaldialdehyde (OPA) was used in this study. The values of analytical blank for individual amino acid varied from 0 to 1.8×10^-3μmol/L, Limits of detection (3σ) ranged from 2.0×10^-3 to 9.0×10^-3. Recoveries of urine added to mixed amino acids standard were between 99.5 % and 99.7%. The relative standard deviation was 3.4%-10.7%. 16 amino acids were detected in the marine colloid. The concentration of THAA was 150.83μmol/L. Lysine was the most abundant compound which concentration was 35.59μmol/L. Its contribution to THAA was 23.6 %. Aspartic acid, glutamic acid, histidine,glycine + threonine, methionine and leucine were the following 7 abundant amino acids. The individual concentration was 12.40, 10.48, 17, 77,16. 60, 17.22 and 15.88 μmol/L respectively and contribution to THAA was 8, 2% ,7, 0% ,11.8% ,11.0%, 11.4% and 10.5% respectively. Together these 8 compounds contributed up to 83.5 % to THAA. The other 8 amino acids contributed only 16.5 %.
