大鼠生后不同发育阶段海马CA_3区PCNA和bFGF表达的研究 被引量:1

摘要 目的:通过观察大鼠生后不同发育阶段海马CA3区PCNA和bFGF的表达,探讨海马的发育规律。方法:取生后第1天、第2天、第3天、第5天、第10天、第20天、第30天大鼠脑,免疫组化染色,观察PCNA、bFGF在海马CA3区的表达情况。结果:大鼠海马CA3区细胞增殖以生后第2天最为明显,其中多形层细胞增殖数量最多,其次是分子层,锥体层细胞增殖最少;第10天以后增殖逐渐减少,第30天时增殖基本停止。bFGF在生后第1~2天时未见表达,第3天开始出现表达,其中以锥体层、多形层细胞阳性表达最多,第20天时表达基本稳定。结论:在大鼠生后海马发育过程中,bFGF的表达对PCNA的表达有促进作用。 Objective.By studying on the expression of PCNA and bFGF in CA3 area of hippocampus to make out the developmental law of hippocampus. Methods: The brain of rats were taken out at postnatal d1, d2, d3, d5, d10, d20, d30 respectively and fixed with 10 % formalin, SP imrnunohistochemistry was used in this study to observe the expression of PCNA, bFGF in CA3 area of hippocampus . Results: The PCNA expression in CA3 area of hippocarnpus was the most obvious at postnatal d2, neurons in multiform layer proliferated the most, multiform layer took second place and pyramidal layer the least. The PCNA expression decreased gradually after postnatal d10 and basically ended at postnatal d30. bFGF positive cells were not observed in CA3 area of hippocampus at postnatal d1-d2. The expression of bFGF reached the top at postnatal d3 and expressed the most in pyramidal layer, and became stable at postnatal d20. Conclusions: In the development of hippocampus, bFGF can stimulate the expression of PCNA.
