摘要
目的:观察并比较Mc9204(C_(20)H_(24)ClNO_4,377)和硝苯地平(Nif)对肾性高血压大鼠血压及肾脏血流动力学的影响.方法:采用颈动脉插管测定血压,比色法测定肾血流量(RBF),肾小球滤过率(GFR)和尿Na ̄+.结果:iv连续输入Mc92041,3,10μg/(kg·min)三个剂量,均能产生明显的降压作用,与空白对照组有显著差异(降压百分比分别为17.73±5.63,36.21±9.00,50.23±5.17us4.22±1.51,5.00±3.07,5.50±3.04,n=7,P<0.01).并表现出剂量依赖性,Mc92041μg/(kg·min)使RBF和GFR同步增加,并出现排钠利尿作用.大剂量时,由于血压的过度降低,此作用被抵消.结论:Mc9204具有显著的降压及增加RBF的作用,此作用明显大于Nif.
Objective:To compare the effects of Mc9204 and nifedipine(Nif)on blood pressure and renal hemodynamics in renal hypertensive rats.Methods:Carotid artery catheter was used to monitor mean blood pressure,and colorimetric assay was used for determination of renal blood flow(RBF),glornerular filtration rate(GFR)and urine Na ̄+. Results:Results showed that Mc9204 iv,1,3,10 μg/(kg·min)could decrease blood pressure in concentration-dependent manner,and presented a significant difference compared with con-trol group(percentages of reduced blood pressure were 17.73±5.63,36.21±9.00,50.23±5.17 us 4.22±1.51,5.00±3.07,5.50±3.04 respectively,n=7,P<0.01).At the concentration of 1μg/(kg·min),it could simultaneously increase RBF and GFR,accompanied by excretion of urine Na ̄+ and urine volume rise.But the effects were attenuated due to overreduced blood pressure at large doses:3,10μg/(kg·min).Con-clusion:Mc9204 is more effective than Nif in significantly decreasing blood pressure and increasing RBF.
