摘要
目的:进行复方四季青提取物雾化吸入给药的药效学研究。方法:小鼠随机分为空白对照组,阳性药物对照组,复方四季青提取物口服给药高剂量组(2.79 g·kg-1)、中剂量组(1.86 g·kg-1)、低剂量组(0.93 g· kg-1),雾化吸入给药高剂量组(2.79 g·kg-1)、中剂量组(1.86 g·kg-1)、低剂量组(0.93 g·kg-1),采用酚红祛痰法观察祛痰作用和体内抗菌法观察对肺炎双球菌感染小鼠的保护作用。结果:复方四季青提取物高剂量口服给药,高剂量雾化吸入、中剂量雾化吸入能使小鼠气管酚红排泌量明显增加,排泌量分别为(0.052±0.009), (0.055±0.009),(0.052±0.009)mg·L-1,有显著的祛痰作用,与空白对照组相比有显著差异(P<0.05)。雾化吸入给药及高剂量口服给药对腹腔感染肺炎双球菌小鼠的死亡率均有明显的抑制作用,72 h死亡率分别为 40%,40%,45%,55%,1 wk死亡率分别为40%,50%,55%,60%。与空白对照组相比有显著差异(P< 0.05)。结论:复方四季青提取物雾化吸入给药是提高疗效的有效途径之一。
AIM:To evaluate pharmacodynamic effects of the extract of compound Chinese holly leaf administrated by aerosol inhalation. METHODS :The rats were randomized into normal control group ,positive control group,high dose gastric ravage group (2.79 g·kg^-1),middle dose gastric ravage group (1.86 g·kg^-1) ,low dose gastric ravage group (0.93 g·kg^-1),high dose aerosol inhalation group (2.79 g·kg^-1),middle dose aerosol inhalation group (1.86 g·kg^-1) and low dose aerosol inhalation group (0.93 g·kg^-1).The protective effects against the infection of Streptococcus pneumoniae were observed by phenol red secretion test and antibacterial test in vivo. RESULTS: Compare with normal control group ((0.043 ± 0.009) mg·L^-1) , the phenol red secretion increased significantly (P 〈 0.05)in high dose gastric ravage group ((0.052 ± 0.009) mg·L^-1) ,high dose aerosol inhalation group ((0.055 ± 0.009) mg·L^-1)and middle dose aerosol inhalation group ((0.052 ± 0.009) mg·L^-1).The mortality decreased significantly(P 〈 0.05)in aerosol inhalation group (40 % ,40 % ,45 % after 72 h and 40 %, 50 % ,55 % after 1 wk respectively)and high dose gastric ravage group(55 % after 72 h and 60 % after 1 wk).CONCLUSION : Aerosol inhalation can improve the therapeutic effect of the compound Chinese holly leaf.
