摘要
对分离自中国南方的H9N2亚型禽流感病毒A/Chicken/Guangxi/KMⅢ/99(H9N2)的M基因进行克隆和序列分析。序列分析表明,该M基因全长1 027 bp,包含2个读码框,可分别翻译出M1和M22种蛋白质。M1由252个氨基酸残基组成,M2由97个氨基酸残基组成。与不同毒株比较,结果显示,该M基因与A/Chicken/Guangxi/10/99(H9N2)同源性为100%,当A/Chicken/Guangxi/9/99(H9N2)同源性为99%,与A/Chicken/Guangdong/5/97(H9N2),A/Chicken/Guangdong/11/97(H9N2)等中国分离株同源性达98%。
According to the reported sequences of M gene from Avian influenza virus(AIV) ,a pair of primers was designed for amplification of M gene of A/chicken/Guangxi/KMⅢ/99 (HgN2) by RT-PCR. The PCR products were cloned into plasmid pMD18-T. The sequence analysis of the insert fragments in the recombinant plasmids pMD-M indicated that the M gene(1027 bp) share more than 100% ,99% ,98% identi ties compared respectively with AIV strains that GenBank show. M gene encodes two proteins: M1 protein and M2 protein. The M1 protein is 252 aa long. The M2 protein is 97 aa long. The homology between this amino acid and other HgN2 M1 amino acid in GenBank is above 98% online.
