摘要
目的探讨组织蛋白酶B(CB)在人胶质瘤中的表达及其与胶质瘤恶性程度、血管生成的关系。方法应用免疫组化(SABC染色)方法检测47例手术切除的星形细胞胶质瘤标本和5例自发性脑内出血手术活检并被证实为正常脑组织标本中CB的表达和微血管密度(MVD)。结果5例正常脑组织CB表达为阴性。星形细胞瘤、间变型星形细胞瘤、胶质母细胞瘤中CB的表达分别为30%(6/20弱阳性),86.7%(3/15弱阳性,10/15阳性),100.0%(2/12阳性,10/12强阳性)。CB染色强度与肿瘤恶性程度相关(r=0.77,P<0.05)。在CB染色呈强阳性、阳性、弱阳性、阴性的标本中,MVD分别为49.90±11.30,39.85±8.75,22.50±14.42,15.57±8.52。CB表达水平与MVD相关(r=0.56,P<0.05)。结论CB表达水平与胶质瘤恶性程度和血管生成密切相关,可能是恶性程度及侵袭性的标志之一。
Objective To test the expression of Cathepsin B (CB) protease in gliomas and to investigate its correlation with the malignance and angiogenesis of gliomas. Methods Immunochemistry technique of strept actividin-biotin complex (SABC) was used to identify the CB expression and to measure the microvessel density (MVD) in 47 gliomas samples and 5 normal human brain tissues from patients with spontaneous cerebral hemorrhage. Results No CB expression was detected in the normal brain tissues. The expression levels of CB in astrocytoma, anaplastoma, and glioblastoma were 30% (6/20 weak positive), 86. 7%(3/15 weak and, 10/15 moderate positive), and 100. 0%(2/12 moderate and 10/12 strong positive), respectively. The malignances of gliomas increased with CB expression (r=0. 77, P〈O. 051. The MVD also increased with CB expression (r= 0.56, P〈0.051, with an average of 15. 57±8. 52, 22. 50±14. 42, 39. 85±8. 75 and 49. 90±11.30 for the negative, weak, moderate, and strong CB expression gliomas respectively. Conclusion CB expression is positively correlated with the malignances and angiogenesis of gliomas. CB expression could be one of the indicators for the degree of malignances and invasion of gliomas.
