摘要 目的研究IL-6对脐静脉血管内皮细胞(HUVECs)组织因子(TF)的mRNA水平的影响。方法应用胰酶消化HUVECs并进行传代培养,用第二、三代细胞进行试验。测定IL-6(0.5ng/mL)处理前后细胞活性,反转录聚合酶链反应(RT-PCR)法检测细胞内TFmRNA水平。结果72h内,IL-6对HUVECs的活性的影响与正常对照组相比差异无统计学意义。IL-6作用HUVECs6h即诱导TFmRNA表达,12h达到高峰,以后表达减弱,72h不能检测到mRNA;对照组TFmRNA不表达或表达极弱。结论IL-6(0.5ng/mL)对HUVECs的活性不造成直接的影响,同时在24~48h内IL-6(0.5ng/mL)可诱导HUVECsTF表达,这可能与IL-6作为前炎症因子诱导急性炎症反应时相的血液循环障碍相关。 Objective To investigate the effects of IL-6 at the expression of tissue factor (TF) in human vein endothelial cells(HUVECs). Methods HUVECs were incubated with IL-6 at the concentration of 0. 5 ng/ mL. Cell viability was measured by CCK-8 assay. The TF mRNA was detected by reverse transcript-polymerase chain reaction(RT-PCR) method. Results When HUVECs were exposed to IL-6(0. 5 ng/mL) within a period of 72 h, their viability did not decrease in comparison with the control; there was no statistical difference between the two groups. After the HUVECs were exposed to IL-6(0.5 ng/mL) for 6 h, the TF mRNA level increased, and it reached the peak at 12 h; then it began to decline. The expression of TF mRNA induced by IL-6 was evidently detected from 6 h to 48 h. After the HUVECs were treated by IL-6 over 72 h, the expression of TF mRNA was no longer detected in HUVECs. Conclusion IL-6 at the concentration of 0. 5 ng/mL did not exert direct effect on cell viability. The increase of TF mRNA expression in HUVECs induced by IL-6 could play an important role in the modulation of blood coagulation disorder and in the mechanism related to coagulation system changes during inflammatory reaction.
