施万细胞和神经干细胞在PLGA支架中共培养的扫描电镜观察 被引量:16

摘要 目的扫描电镜观察施万细胞和神经干细胞在生物可降解材料(乙交酯-丙交酯)共聚物(Poly(lactide-co-glycolide acid),PLGA)支架中的生长状态,探讨施万细胞、神经干细胞与 PLGA取向支架的细胞亲和性。方法体外培养大鼠神经干细胞和施万细胞,待细胞生长良好,移植接种至PLGA支架上,培养9d后行扫描电镜观察。结果施万细胞与神经干细胞在PLGA支架表面贴附,形态正常,生长良好。迁出神经球的神经干细胞分化成多种形态的细胞紧贴PLGA表面, 部分细胞形态类似神经细胞。结论施万细胞和神经干细胞在PLGA支架中生长良好,PLGA对施万细胞及神经干细胞具有良好的细胞亲和性。 Objective To study morphology of Schwann cells and neural stem cells cultured on biodegradable Poly (lactide-co-glycolide acid) (PLGA) scaffold with scanning electron microscopy (SEM) and investigate the affinity of Schwann cells and neural stem cells to PLGA scaffold. Methods Schwann cells and neural stem cells were cultured in vitro, and then transseeded into PLGA scaffold to culture for 9 days. The cell morphology was investigated by SEM. Results These cells grew in adherent way on the surface of PLGA scaffold and had normal shapes. Some differentiated neural stem cells emigrated out of neurosphere. The shape of some cells was similar to that of the neurons. Conclusion. The growth of these cells on the PLGA scaffold is normal. The affinity of PLGA scaffold to Schwann cells and neural stem cells is good.
