摘要
目的:观察反映端粒酶的活性的端粒酶催化亚单位在病理性瘢痕组织中的表达情况。方法:实验于2005-05/07在长海医院中心实验室进行。取增生性瘢痕标本和瘢痕疙瘩标本各18例,来源于2004-06/2005-05解放军第二军医大学长海医院整形外科手术患者;正常皮肤标本18例,取自瘢痕邻近的正常皮肤。采用SP免疫组化法,对3组皮肤标本中成纤维细胞端粒酶催化亚单位蛋白表达进行检测,观察其阳性颗粒的平均吸光度A值和阳性面积率。结果:①端粒酶催化亚单位阳性颗粒的平均吸光度:瘢痕疙瘩组高于正常皮肤和增生性瘢痕组(5.940±0.675,0.456±0.078,1.352±0.193,P<0.01),增生性瘢痕组高于正常皮肤(P<0.05)。②端粒酶催化亚单位的阳性面积率:瘢痕疙瘩组高于正常皮肤和增生性瘢痕组眼(0.203±0.025)%,(0.038±0.010)%,(0.067±0.011)%,P<0.01演,增生性瘢痕组高于正常皮肤(P<0.05)。结论:端粒酶在病理性瘢痕中表达增高,与病理性瘢痕发生、发展及其演变过程密切相关。设想减少端粒酶催化亚单位在病理性瘢痕成纤维细胞的过度表达从而抑制端粒酶活性或许是抑制瘢痕增生的新途径。
AIM: To observe the expression of telomerase catalytic subunit, which can reflect telomerase activity, in pathological scar tissues. METHODS: The experiment was undertaken in the Central Laboratory of Changhai Hospital from May to July 2005. Samples of hyperplastic scar (n=18) and keloid (n=18) were taken from patients in the Department of Plastic Surgery, Changhai Hospital of Second Military Medical University of PLA from June 2004 to May 2005; Normal skin samples (n=18) were taken from normal skins closed to scars. Protein expressions of fibroblasts telomerase catalytic subunit in samples of three groups were inspected with SP immunohistochemical method. Average absorbance A and positive area ratio of positive granules were observed. RESULTS: ①Average absorbtance of telomerase catalytic subunit in positive granules: That in the keloid group was higher than that in normal skins and hyperplastic scar group (5.940±0.675,0.456±0.078,1.352±0.193 ,P 〈 0.01 )and that in hyperplastic scar group was higher than that in normal skins (P 〈 0.05). ②Positive area ratio of telomerase catalytic subunit: That in the keloid group was higher than that in normal skins and hyperplastic scar group [(0.203±0.025)%, (0.038±0.010)%, (0.067±0.011)% ,P 〈 0.01] and that in the hyperplastic scar group was higher than that in normal skins ( P 〈 0.05). CONCLUSION: Expression of telomerase in pathological scars increased and in close relation to the pathogensis, development and variation process of pathological scars. Inhibition on overexpression of fibroblasts in telomerase catalytic subunit of pathological scars may be the new method of inhibiting telomerase activity or proliferation of scars.
