摘要
目的研究不同剂量的氟对体外培养大鼠乳鼠成骨细胞中Ⅰ型胶原表达的影响。方法体外培养Wistar大鼠乳鼠成骨细胞,给予不同剂量的氟,提取RNA,采用逆转录聚合酶链反应(RT-PCR)方法检测Ⅰ型胶原mRNA:采用酶联免疫吸附实验(ELISA)方法检测培养液上清中的Ⅰ型胶原蛋白质。结果与对照组相比,1、2及4 mg/L F-均能促进Ⅰ型胶原表达,但在蛋白质表达方面培养48 h后只有2、4 mg/L F-组与正常对照组差异有统计学意义(P<0.05),而在培养72 h后各加氟组与正常对照组相比差异均有统计学意义(P< 0.01);在mRNA表达方面培养48 h后只有2 mg/L F-组与正常对照组差异有统计学意义(P<0.01),而在培养 72 h后2 mg/L F-和4 mg/L F-组与正常对照组相比差异均有统计学意义(P<0.01)。结论氟可促进成骨细胞Ⅰ型胶原的表达,且与氟化物水平以及作用时间有关。
Objective To study the influence of fluoride on the expression of collagen type Ⅰ (Col Ⅰ ) in rat osteoblasts. Methods Osteoblasts obtained from calvaria of sucking Wistar rats were cultured in medium with different concentrations of NaF. The effects of different concentrations of NaF on the expression of Col Ⅰ mRNA expression were determined by RT-PCR and protein expression were evaluated by ELISA respectively. Results 1, 2 and 4 mg/L F^- all promoted the expression of Col Ⅰ at mRNA and protein level, but only 2 and 4 mg/L F^- exerted significant influences on the expression of protein (P 〈 0.05) compared to the control group at 48 h, while 1, 2 and 4 mg/L F^- all had such influences (P 〈 0.01) at 72 h; only 2 mg/L F^- had significant influences on the expression of mRNA (P 〈 0.01 ) compared to the control group at 48 h and 2 and 4 mg/L F^- at 72 h (P 〈 0.01 ). Conclusions The results suggest that NaF might promote the expression of Col Ⅰ in sucking rat osteoblasts according to the concentrations of NaF and reacting time.
