摘要
目的探讨自分泌运动因子Autotaxin(ATX)在人卵巢癌中的表达及其与卵巢癌彩色血管能量成像(CPA)特征之间的关系。方法术前应用经阴道CPA检查,术后应用半定量逆转录聚合酶链反应(RT-PCR)、Western blot印迹研究8例正常卵巢组织、18例良性卵巢上皮性肿瘤、50例卵巢上皮性癌组织、23例大网膜转移灶中ATX mRNA的表达,以-βactin为内参照。结果正常卵巢、卵巢良性肿瘤、卵巢癌、大网膜转移灶中均有ATX mRNA不同程度表达;但ATX基因表达值癌组织和大网膜转移灶中显著高于良性卵巢肿瘤和正常卵巢组织[分别为(89.31±10.15)%(、88.91±11.05)%(、40.18±16.71)%、(35.29±13.82)%,P<0.001]。Western blot显示ATX蛋白在卵巢癌细胞中明显表达,在55 kD6、0 kD大小的位置可见清楚显色的条带。ATXmRNA在卵巢癌细胞亦可见明显扩增条带。说明卵巢癌细胞中ATX在核酸与蛋白水平均有表达,两者结果一致。高表达ATX卵巢癌与癌细胞转移、癌栓形成、肿瘤的分化程度有关,而与年龄、肿瘤大小等无关。卵巢癌CPAⅢ型病例的ATX表达明显高于Ⅰ型和Ⅱ型病例,实性区少的上皮性癌乳头区ATX表达最高。结论卵巢癌细胞中ATX分子在核酸与蛋白水平均有表达,两者结果一致;CPA为临床鉴别良恶性卵巢肿瘤提供了敏感的血流信息,且CPA血流特性与ATX基因、蛋白表达存在正相关,ATX过度表达与卵巢癌的进展、转移有关。
Objective To investigate the expression of autotaxin(ATX)mRNA and protein in the human ovarian epithelial carcinoma and whether there is correlation between ATX expression and color power anglo imaging(CPA) features of ovarian epithelial carcinoma. Methods Eight specimens of normal ovarian tissues, 18 cases with ovarian epithelial benign tumor,50 cases with ovarian epithelial carcinoma and 23 cases with metastatic focus of omentum were studied. Semi-quantitative reverse transcriptionase-polymerase chain reaction(RT-PCR) and Western blot analysis were employed to detect mRNA and protein expression of ATX. Results ATX protein and mRNA was expressed,the expression level of ATX gene in ovarian epithelial carcinoma and metastatic focus of omentum were higher than those in ovarian epithelial benign tumor and normal ovarian tissues [(89.31 ± 10. 15)% ,(88.91 ± 11.05)%, (40. 18 ± 16.71)%, (35.29 ± 13.82)%, respectively, P〈0. 001], Western blot amino acid analysis demonstrated that ATX was higher expression in ovarian carcinoma than normal ovarian tissues,clearly ladder was in the position 55 kD, 60 kD, ATX mRNA also had clearly expression in ovarian carcinoma. The result corresponded to protein expression. Metastasis, vascular invasion and poor differentiation were more frequently noted in ovarian carcinoma with high ATX expression than in patients with low ATX expression. ATX expression in ovarian carcinoma with CPA typeⅢ were obviously higher than those in type Ⅰ and typeⅡ , specially papillary regional ATX expression were highest. Conclusions The expression of ATX gene corresponds to the protein expression in ovarian epithelial carcinoma,CPA provides sensitive flow information for differentiating epithelial ovarian tumors, CPA flow features corresponds to ATX gene and protein expression of ovarian epithelial carcinoma. High ATX expression are related to metastasis vascular invasion and poor differentiation.
