摘要
目的 观察生脉注射液对5-FU抗肿瘤增效减毒作用。方法 建立H22肝癌荷瘤鼠模型后,随机分为5组:对照组、5-FU组、生脉注射液(大、中、小剂量)+5-FU组,每组10只。造模次日开始进行干预,对照组、5-FU组、生脉注射液(大、中、小剂量)+5-FU组分别腹腔注射等容量生理盐水、5-FU、生脉注射液(大、中、小剂量)+5-FU,用药14d。停药次日处死实验小鼠,观察瘤重抑制率、免疫功能、肝肾功能、外周血细胞变化。结果 ①生脉注射液联合5-FU各组肿瘤生长明显低于5-FU组和对照组(P〈0.05);②与对照组相比,5-FU组CD3^+、CD4^+、CD4^+/CD8^+、IgG、IgM值明显降低(P〈0.05),而各生脉联合组CD3^+、CIM^+、CD4^+/CD8^+.IgG、IgM值明显升高(P〈0.05);③与对照组相比,5-FU组ALT升高、WBC和PLT降低,而各生脉联合组相对于5-FU组ALT降低、WBC和PLT升高(P〈0.05)。结论 生脉注射液可增加5-FU的抑瘤效果,提高免疫功能,减少化疗毒副反应。
Objective To observe the effect of Shengmai injection on enhancing efficacy and reducing toxicity of chemotherapeutic agent, 5-FU. Methods Hepatoma 22 bearing mice models were established and then divided randomly into five groups, including control group,5-Fu group, Shengmai injection (low, middle and high dose) combined with 5-FU groups. There were 10 mice in each group. The five groups were injected introperitoneally with same amount of natural saline, 5-FU 20mg/kg and Shengmai injection (3.5mL/kg, 7mL/kg and 14mL/kg) plus 5-FU, respectively, once a day for 14 days. After that, killed and observed the tumor inhibiting rates and the index of immunological function, liver and kidney function and the blood cell in the peripheral blood. Result ①The tumor inhibiting rates were higher in each Shengmai injection plus 5-FU group than that in the 5-FU group (P〈0.05); ②Compared with the control group, the level of CD3, CD4, CD4/ CD8, IgG and IgM in 5-FU group were lower (P〈0.05). But in the three Shengmai injection groups combined with 5-FU they were higher than that in control group and 5-FU group, which revealed significant difference (P〈0.05). ③The level of serum ALT were higher in 5-FU group with lower WBC and PLT counts in the peripheral blood. But, in the three Shengmai injection plus 5-FU groups, the level of serum ALT were lower and WBC and PLT counts were higher than that in the 5-FU group (P〈0.05). Conclusion Shengmai injection can enhance the effect of anti-tumor efficacy of 5-FU. It can also improve the immunological function and reduce the side-effect of chemotherapy.
