摘要
葛根素对胰蛋白酶有明显的抑制作用,D ixon作图法得出抑制常数Ki=8.15×10-5mol.L-1,抑制类型为非竞争性抑制。通过紫外吸收光谱,发现葛根素能改变胰蛋白酶的空间结构;由荧光光谱变化探索葛根素与胰蛋白酶的相互作用机理,发现荧光猝灭是能量转移引起的静态猝灭。
The prohibitive function of puerarin over trypsin was studied. The value of Ki (8. 15 × 10^-5mol/L) was obtained by using Dixon graphical construction according to the ratio between 1/v and amount of inhibitor. It was found that inhibitor was a noncompetitive. Utilizing ultraviolet spectroscopy,we found that puerarin can alter the molecular conformation of the trypsin. Utilizing fluorescence spectrophotometry we studied the mechanism of the interaction between puerarin and trypsin and found that fluorescence quenching was a static quenching due to nonradiative energy - transfer.
