精索静脉曲张大鼠睾丸组织HIF-1α、VEGFi、NOS的表达及意义 被引量:3

摘要 目的:检测HIF-1α、VEGF及iNOS在实验性精索静脉曲张(VC)大鼠睾丸组织中的表达,探讨精索静脉曲张导致男性生育力低下的病理生理学机制。方法:用SD雄鼠制造VC模型。36只SD雄性大鼠分为对照组(n=10)、假手术组(n=11)和VC组(n=15),造模后30天行左侧睾丸切除,用免疫组织化学方法检测睾丸组织中低氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)及诱导型一氧化氮合酶(iNOS)的表达。结果:HIF-1α、VEGF和iNOS在VC组睾丸组织中的表达均上调,明显高于对照组和假手术组(P<0.05)。结论:青春期实验性VC大鼠的左侧睾丸组织存在低氧。低氧引起HIF表达并激活其下游基因表达,是精索静脉曲张一系列病理生理学改变的可能基础。NO表达上调与间质细胞中iNOS的激活有关。 Objective: In this study we investigated hypoxia inducible factor-1α, vascular endothelial growth factor and inducible nitric oxide synthase (iNOS) expression in an experimental model of varicocele in the rat testis and try to find out the pathophysiology of varieoeele in male infertility. Methods: A total of 36 adult male SpragueDawley rats were investigated in 3 groups, namely varieoeele group 1 (15), sham operation group 2 (11) and con trol group 3 (10). At 30 days after surgery was completed, orchiectomy was performed. Histological findings in the left testicles of rats from 3 different groups were compared with each other. Results:The expression of HIF-1α, VEGF and iNOS in left testes is significantly higher than that of control or sham-operation group. Conclusions:The results of this study show that varieocele can lead to local tissue hypoxia in left testis and activate it's target gene. Maybe this is the reason that varicocele can damage the testes.
