胃癌gp96多肽复合物树突状细胞疫苗对Th1/Th2平衡的影响 被引量:1

摘要 目的探讨gp96多肽复合物树突状细胞(dendriticcells,DCs)疫苗对Th1/Th2平衡的影响。方法从胃癌组织中提取gp96多肽复合物负载从外周血单个核细胞诱导培养的DCs,用gp96多肽复合物DC疫苗诱导同一患者Th0淋巴细胞,ELISA检测培养上清中IFNγ和IL10的水平;以胃癌细胞裂解物负载的DC疫苗作对照。结果gp96多肽复合物DC疫苗诱导的T细胞分泌IFNγ的量为(2875±177.66)pg/ml,显著高于对照组(1765±289.07)pg/ml(P<0.01);分泌IL10的量为(36±6.72)pg/ml,显著低于对照组(90±11.26)pg/ml(P<0.05),二者之间存在显著的负相关(r=-0.915,P<0.01)。结论gp96多肽复合物DC疫苗可诱导Th1/Th2平衡向Th1漂移,显示了gp96多肽复合物DC疫苗可显著提高机体的免疫能力,为临床免疫干预治疗奠定基础。 Objective To study the effect of gp96 polypeptide complex-dendritic cell vaccine on Th1/Th2 balance. Methods gp96 polypeptide complex was purified from gastric cancer tissues and eoeuhured with mononuelear cells derived from peripheral blood. DCs from above process were mixed with autogenie Th0 lymphoeytes and the level of IL-10 and IFN-γin supernant was determined using ELASA. DC loaded with autologous gastric cancer cells lysates was as control. Results The level of IFN-γ secreted by T lymphoeytes induced by DCs vaccine loaded with gp96 polypeptide complex was (2 875 ± 177.66) pg/ml, higher than ( 1 765 ±289.07) pg/ml of control (P 〈0.01 ), and the level of IL-10 was (36 ± 6.72) pg/ml, lower than (90 ± 11.26 ) pg/ml of control ( P 〈 0.05 ), suggesting a negative correlation between IL-10 and IFN-γby linear correlation analysis ( r = - 0. 915, P 〈 0.01 ). Conclusion gp96 polypeptide complex-dendritic cell vaccine could induce Th0 differentiate to Thl, indicating its capability against tumuor.
