NGF和神经节苷脂GM_1对2,5-己二酮中毒性PC12细胞的协同保护作用 被引量:4

摘要 目的:研究神经生长因子(Nerve Growth Fac-tor,NGF)和神经节苷脂GM1(Ganglioside GM1)对2,5-己二酮(2,5-HD)中毒的保护作用。方法:将细胞随机分为对照组和实验组,对照组于正常培养基生长,实验组以2,5-HD、NGF和GM1处理。采用Giemsa染色法在倒置显微镜下观察细胞形态,MTT法检测细胞存活率,流式细胞计数法检测细胞凋亡率。结果:2,5-HD处理后,细胞损伤存活率降低(P<0.01),凋亡率上升(P<0.01);以NGF和GM1预处理的细胞显示出明显的抗损伤性,细胞存活率提高(P<0.05),凋亡率下降(P<0.05),且在联合时较单独应用效果更显著(P<0.01)。结论:2,5-HD抑制PC12细胞存活和增殖,诱导细损伤和凋亡。NGF和GM1对2,5-HD中毒性PC12细胞显示出协同保护作用。 AIM: To investigate whether NGF and GM1 have synerg/stic protective effect on 2,5-HD intoxication in PC12 cells. METHODS: Cells were divided into control group and treated groups at random. Control group was maintained in normal medium; treated groups were exposed to 2,5-HD with or without NGF, GM1. The cells' morphological changes were observed by inverted microscope after being dyed with Giemsa. The cell viability was determined by MTT assay. The apoptotic ratio was determined by flow cytometry (FCM). RESULTS:PC12 cells which was treated by 2,5-HD injury aggravated, the cell viability decreased ( P 〈 0.01 ), and the apptotic ratio increased (P 〈 0.01 ). PC12 cells pretreated with NGF and GM1 obviously resisted injury, the cell viability increased ( P 〈 0.05), apoptotic ratio decreased ( P 〈 0.05). Combination of NGF and GM1 afforded additional protection that was greater than that of either agent individually ( P 〈 0.01). CONCLUSION: 2,5-HD inhibited the survival, induced apoptosis and injury of PC12 cells. NGF and GM1 showed synergistic protective effect on 2,5-HD intoxication in PC12 cells.
