摘要
利用陆地棉EST—SSRs(Expressed sequence tags—simple sequence repeats)在陆地棉和向日葵品种中进行了通用性分析。利用本实验室合成的1500对陆地棉EST—SSRs在4个陆地棉品种中进行了扩增,有效扩增比率为95%,扩增产物符合预期大小(50~500bp);每对引物可以检测到1~6个数目不等的片段(allele),平均约为2.7个;每2个陆地棉之间大约有7%~9%的多态性。从1500对陆地棉EST—SSRs引物中选取400对引物在向日葵中进行了扩增,有效扩增比率为63%.每对引物可以检测到1~6个数日不等的片段,平均为1.7个,片段大小介于50~500bp之间。选取了200对在向日葵中得到有效扩增的引物对向日葵G101的父母本进行了多态性筛选,约25%引物具有多态性。结果表明,陆地棉EST—SSRs在向日葵中具有较高的通用性,可用于向日葵比较基因组研究和分子标记研究。
Transferability ot upland cotton EST-SSRs to upland cotton ancl sunflower were anaiyzea. 1500 pairs of upland EST-SSRs synthesized in the study were amplified in four varieties of upland cotton,their effective amplification percentage reaching 95% and their amplification products being what were expected ;each pair of primers could be used to test a certain number of bands which varied within 1 -6 and averaged at 2. 7;in every two varieties of upland cotton polymorphic bands occupied 7%-9%;400 pairs of primers chosen from the 1 500 pairs of primers were amplified in sunflower,their effective amplification percentage standing at 63% ,each pair of primers capable of testing a certain number of bands which ranged within 1-6 and averaged at 1.7,and whose sizes ranged within 50-500 bp. 200 pairs of primers that could effectively amplified in sunflower were chosen to test their polymorphisms in the parents of G101 ,finding that about 25% of them were polymorphic. The results showed that upland cotton EST-SSRs had high transferability to sunflower and thus they can be used in comparative genome and molecular marker researches.
