不同温度保存对骨髓细胞存活率的影响

摘要 目的探讨4℃与-196℃保存骨髓细胞的方法和存活率。方法取wistar大鼠,无菌条件下制备成骨髓细胞悬液,分别于4℃与-196℃保存,保存96 h和60 d后,用Trypanblue染色、观察骨髓细胞的回收率。结果骨髓细胞4℃保存96 h,回收率为28;-196℃保存60 d,细胞回收率为2318。结论-196℃保存骨髓细胞其存活率高于4℃保存,它是中长期保存骨髓细胞的一种有效的方法。 Objective To probe into surviving rats of preservation bone marrow cells in 4℃ and - 196 ℃. Methods The authors took bone marrow cells of naked rats and made marrow cells with trypanblue dyeing af- ter preservation 96 h and 60d, Results The retrieve rate of bone marrow cells was 28 in preservation 96 h of 4 ℃. It is 2318 in preservation 60 d of - 196 ℃. Conclusion The retrieve rate of preservation with - 196 ℃ is higher than preservation with 4 ℃ ( P 〈 0.05). It is an effective preservation method of a middle and long period of time preservation bone marrow cells with - 196 ℃.
