摘要
以野生马利筋二倍体种子为材料,在离体培养条件下,采用不同浓度秋水仙素(0.03%~0.10%),在不同的处理时间条件下(24~72h)进行诱导。试验表明,在0.10%浓度,48h条件下诱导效果最好。首次鉴定出了二倍体马利筋染色体数目为2n=2×=20,诱导出的同源四倍体马利筋染色体数目为2n=4×=40。四倍体与二倍体在形态学和细胞学上有明显差异。
The wild diploid seeds of Asclepias curassavica are used as material. The material is induced with different concentration colchicine (0. 03 % - 0. 10% ) and different time ( 24 - 72 h) on the condition of tissure culture. The result shows that on the condition of 0. 10% and 48 h,the effects are best. And it is the first time reported that the chromosome number of the diploids is 20, and the chromosome number of autotetrapIoid is 40. There are obvious difference on morphology and cytology between polypoid and diploid.
