Effect of IL-1RA on keratocyte collagen degradation by pseudomonas aeruginosa

目的:探讨白细胞介素1受体拮抗剂(IL-1RA)在绿脓杆菌引起的角膜基质细胞胶原降解中的作用,为临床上绿脓杆菌性角膜炎的治疗提供新的理论依据。方法:角膜基质细胞胶原凝胶及无角膜基质细胞胶原凝胶与绿脓杆菌培养液共同培养24h,测定培养上清液中羟脯氨酸的含量。同时检测IL-1RA对胶原降解的抑制作用。结果:绿脓杆菌培养液能直接降解Ι型胶原,在角膜细胞存在的情况下降解作用更强。角膜细胞存在时,IL-1RA可以明显抑制绿脓杆菌所致的胶原降解作用。结论:IL-1RA具有抑制绿脓杆菌引起的角膜基质细胞胶原降解的作用。

Objectlve To study the effect of IL-IRA on keratocyte collagen degradation by pseudomonas aeruginosa for instruction of clinical treatment to pseudomonas aeruginosa keratitis. Method Collagen gels with or without suspended keratocytes were incubated for 24 hours under medium containing sterile pseudomonas aeruginosa culture broth,and the amount of hydroxyprline of the culture supernatants were measured.The inhibitive effect of IL-1RA on collagen degradation were also examined. Results pseudomonas aeruginosa broth induced collagen gel degradation direcdy.ln the presence of keratocytes,the degradation was enhanced IL-1RA significandy suppressed collagen degradation by pseudomonas aeruginosa in the presence of keratocytes. Conclusion IL-1RA inhibites keratocyte collagen degradation by paeruginosa aeruginosa.
