摘要
目的:探讨骨调素(OPN)和单核细胞趋化蛋白(MCP1)在大鼠梗阻性模型中的表达及其在肾脏纤维化发病机制中的作用。方法:采用单侧输尿管结扎(UUO)制造梗阻性肾病模型,分别于造模后第7天、14天取肾组织,用HE和Masson染色法观察UUO大鼠术后不同时间点肾脏病理改变,免疫组化方法检测肾组织中OPN和MCP1蛋白的表达。结果:OPN、MCP1表达主要位于肾小管上皮细胞,随着梗阻时间的延长,肾组织中OPN、MCP1蛋白表达明显增加。表达分别与肾间质胶原相对面积呈正相关。结论:OPN、MCP1蛋白在梗阻性肾病大鼠肾组织表达明显增加介导炎症过程,参与肾间质纤维化。
To investigate the expression and function of OPN and MCP-1 in the kidney after unilateral ureteral obstruction in rats. Methods: Unilateral ureteral obstruction(UUO) models were induced by ligating the left ureter. Rats were sacrificed at the 7th and 14th day after UUO. Interstitial changes were also observed by HE and Masson staining. Immunhistochemistry staining was applied to measure the expression of OPN and MCP-1. Results: In comparison with control group, the expression of OPN and MCP-1 protein were remarkly increased (P〈0.01). The expression of OPN and MCP-1 protein was correlated with the grade of interstitial fibrosis. Conclusion: The protein of OPN and MCP-1 is increased significantly in the obstructed kidney, which may cause renal fibrosis.
