摘要
目的:建立一种简便的巨噬细胞分离与培养方法,为骨替代材料降解产物的研究提供巨噬细胞。方法:以无血清DMEM培养液灌洗小鼠腹腔,10min后吸出灌洗液于含10%胎牛血清的DMEM培养液中培养。采用HE染色和倒置显微镜观察细胞形态;酸性磷酸酶及非特异性酯酶染色测定细胞内酶的表达;墨汁颗粒吞噬实验检测其吞噬功能。结果:获得高纯度的巨噬细胞,具有巨噬细胞的形态特征,良好的吞噬功能,含水解酶类。结论:本法是一种简易而实用的体外分离培养腹腔巨噬细胞的方法。
Objective: To establish a convenient method of separation and cultivation of macrophage. Methods: Abdominal cavity of were douched using DMEM medium without serum, and the contain cells was drawn out and were cultured in DMEM medium with 10% fetal bovine serum. The cells were identified by morphological observation, endocellular enzyme and phagocytosis. Results: The obtained cells were highly purified macrophages with morphologic characteristics of the macrophage in organism, favourable phagocytosis, acid phosphatase (ACP) and α-acetic acid naphthol esterase(α-ANE) activity. Conclusion: This was a simple and pragmatic method for separation and cultivation of mouse peritoneal macrophages.
