苯乙酸钠对肺癌A549细胞生长的影响

摘要 目的:研究抗肿瘤药物苯乙酸钠(NaPA)对体外培养的人肺腺癌细胞株A549抑制率、细胞周期和凋亡的影响。方法:应用四甲基偶氮唑蓝(MTT)比色、流式细胞仪等方法,观察不同浓度NaPA对人肺腺癌A549细胞株生长、细胞周期和凋亡的影响。结果:NaPA对A549有明显的生长抑制作用,且呈剂量时间依赖性,最大抑制率可达79%。NaPA可阻滞A549细胞的细胞周期,使G1/G0期细胞增多,S期细胞减少,同时诱导A549细胞的凋亡,凋亡率最高可达30.5%。结论:苯乙酸钠抑制A549细胞生长,其抑制作用可能是通过NaPA使细胞周期抑制于G1/G0期和诱导细胞凋亡而实现。 Objective: To investigate the effect of sodium phenylacetate (NaPA) on lung adenocarcinoma cell line A549. Methods: Time and dose effects of NaPA were deceted using MTT color metric assay and the influences in the cell cycle and the apoptosis were observed through flow cytometry. Results: NaPA could inhibit the growth of A549 cell line in a dose-depended manner. The biggest rate of cell growth inhibition was up to 79% in the those treated with NaPA. The cytometry confirmed that NaPA could arrest the lung cancer cells from G0/G1 to S stage and induce A549 cells apoptosis. Conclusion: NaPA can inhibit A549 cell line, and the mechanism may be related to the influence in the cell cycle and induction of apoptosis.
