摘要
目的:探讨角质形成细胞中,反义蛋白激酶Cζ与磷脂酰肌醇3激酶(PI3K)和细胞外信号调节激酶ERK1/2信号转导网络以及与核因子NF-κB的相互作用关系。方法:通过细胞原位杂交、间接免疫荧光和激酶活性测定方法,研究反义PKCζ抑制角质形成细胞增殖的信号转导机制。结果:表达反义PKCζ的角质形成细胞中,PI3K的基因表达显著下调,ERK1/2和NF-κB的核内表达水平以及ERK1/2的激酶活性明显降低。结论:表达反义PKCζ可通过下调PI3K和ERK1/2信号通路抑制角质形成细胞的增殖。
Objective: To investigate the inter- reactions between anti - sense protein kinase C (PKC)ξ and the network of PI3K and ERK1/2, and NF -kB in keratinocytes. Methods: The mechanism of signal transduction about the inhibition of anti - sense PKC 4 on the proliferation of Colo16 keratinocytes was measured by cellular hybridization in situ, indirect immunofluoresence and activity assay. Results: The expression of PI3K was decreased notably. The fluorescence intensity of ERK1/2 and NF -kB in nuclear was diminished obviously and ERK1/2 activity was down- regulated in Colo16 keratinocytes expressing anti- sense PKC 4- Conclusion: Anti - sense PKCξ may inhibit the proliferation Colo16 kerationocytes by down- regulating PI3K and ERK1/2 signal pathways.
