摘要
目的建立防风色原酮部位中4种成分升麻素苷、升麻素、5-O-甲基维斯阿米醇苷和亥茅酚苷的高效液相色谱含量测定方法。方法采用Agilent Zorbax C18柱(4.6 mm×150 mm,5μm),甲醇-水梯度洗脱,流速1.0 mL/m in,检测波长254 nm。结果升麻素苷平均回收率为102.56%、102.12%和95.50%,RSD分别为2.24%、1.33%和0.48%(n=5);升麻素平均回收率为95.66%、9.60%和97.54%,RSD分别为0.62%、1.23%和0.89%(n=5);5-O-甲基维斯阿米醇苷平均回收率为98.96%、96.76%和96.91%,RSD分别为0.48%、2.46%和2.74%(n=5);亥茅酚苷平均回收率为99.02%、103.40%和103.96%,RSD分别为2.51%、0.93%和1.61%(n=5)。结论3批样品测定结果表明,该方法简便、准确,可用于防风色原酮部位中升麻素苷等4种成分的含量测定。
Objective To set up a quantitative determination method of HPLC for four components in the chromone fraction of Saposhnikovia divaricata (Turcz.) Schischk, including prin-O-glucosyl- cimifugin, cimifugin, 4'-O-β-D-glucosyl-5-O-methylvisamminol and sec-O-glucosylhamaudol. Methods The determination was carried out with Agilent Zorbax C18 column (4.6 mm × 250 mm, 5 p.m) and gradient elution program of methanol-water. The mobile phase at a flow rate of 1.0 mL/ min and detection wavelength was 254nm. Results The average recovery of prin-O-glucosylcimifugin was respectively 102.56% , 102.12% and 95.50% ( RSD was 2.24% , 1.33% and 0.48% respectively, n = 5 ), that of cimifugin, 95. 66%, 99. 60% and 97. 54% respectively ( RSD, 0.62% , 1. 23% and 0. 89% , n = 5 ) , that of 4'-O-β-D-glucosyl-5-O-methylvisamminol, 98.96%, 96.76% and 96.91% respectively ( RSD, 0.48% , 2.46% and 2.74%, n = 5 ) and that of sec-O-glucosylhamaudol, 99.02%, 103.40% and 103.96% respectively ( RSD, 2.51% , 0.93% and 1. 61%, n = 5). Conclusion The determination results of three batch samples showed that the method was easy, accurate and could be used in the quantitative determination of four components in the chromone fraction of Saposhnikovia divaricata ( Turcz. ) Schischk.
