摘要
采用单因素浓度试验和正交试验研究了Mg2+、Ca2+、Fe2+、Mn2+和Zn2+离子对液体培养霍山石斛类原球茎增殖的影响。结果表明,5种金属离子对类原球茎增殖影响的主次顺序为Fe2+>Mn2+>Mg2+>Ca2+>Zn2+,促进类原球茎增殖的优化组合为Mg2+1.5 mmol.L-1、Ca2+4.5 mmol.L-1、Fe2+0.1mmol.L-1、Mn2+0.5 mmol.L-1、Zn2+0.06 mmol.L-1。经优化的液体培养基增殖的类原球茎能保持植株再生能力,平均每克类原球茎增殖后可再生植株1 985.2株,是相同时间内对照类原球茎再生植株数的4.2倍。
This research is concerned with optimization of Mg^2+ , Ca^2+ , Fe^2+ , Mn^2+ and Zn^2+ for protocorm-like body proliferation in liquid culture of Dendrobium huoshanense by one factor-at-a-time and orthogohal matrix methods. Results showed that, except for Zn^^2+ , the effects of other metal ions on PLB proliferation were significant and the order was Fe^2+ 〉 Mn^2+ 〉 Mg^2+ 〉 Ca^2+ 〉 Zn^2+ The optimal concentration combination for PLB proliferation was determined as 1.5 mmol· L^-1 Mg^2+ , 4. 5 mmol· L^-1 Ca2. , 0. 1 mmol· L^-1 Fe^2+, 0. 5 mmol· L^-1 Mn^2+ and 0. 06 mmol· L^-1 Zn^2+. Proliferated PLBs remained the potential of plant regeneration. The amount of regenerated plantlets from per gram PLBs through proliferation in optimized liquid medium was 1 985.2 and was about 4. 2 times that from PLBs that proliferated on original medium.
