脐血间质干细胞移植对肌萎缩侧索硬化患者血浆和脑脊液中谷氨酸浓度的影响 被引量:7

摘要 目的观察脐血间质干细胞(UCB-MSCs)移植对肌萎缩侧索硬化(ALS)患者血浆及脑脊液谷氨酸(Glu)浓度的影响。方法治疗组8例ALS患者采用静脉滴注结合腰椎脊髓蛛网膜下腔注射UCB-MSCs悬液,行UCB-MSCs移植治疗。应用柱前衍生反相高效液相色谱法分别测定对照组(非神经系统疾病组)和治疗组不同治疗时间血浆和脑脊液中Glu的浓度。结果与治疗前相比,治疗组治疗后3周、3个月血浆、脑脊液Glu浓度降低(P<0.05),但均高于对照组(P<0.05);治疗后3周与3个月相比,治疗组血浆、脑脊液Glu浓度差异无统计学意义(P>0.05)。结论脐血间质干细胞移植治疗能够降低ALS患者血浆、脑脊液中Glu浓度。 Aim :To observe the effect of transplantation of umbilical cord blood mesenchymal stem cells(UCB-MSCs) on glutamate(Glu) level in plasma and CSF of amyotrophic lateral sclerosis (ALS) patients. Methods:Eight ALS patients (treatment group) were treated by intervenous drop infusion and lumbar puncture to transplant UCB-MSCs, and thirteen patients without neural system disease were the control. RP-HPLC were used to detect the change of Glu levels in plasma and CSF of the two groups in different therapic stage. Results: Compared with the control group, the Glu levels in plasma and CSF of treatment group before treatment,one month and three months after treatment were higher ( P 〈 0. 05 ) ; Compared with those before treatment, the Glu levels in plasma and CSF of treatment group one month, three months after treatment decreased significantly(P 〈0. 05 ). There was not significant difference in Glu levels in plasma and CSF of treatment group between those one month and threes months after treatment ( P 〉 0. 05). Conclusion : The treatment of transplant of UCB- MSCs may reduce Glu levels in CSF and plasma of ALS patients.
