摘要
目的研究辛伐他汀对糖皮质激素性骨质疏松的预防作用。方法取6月龄SD雄性大鼠36只,随机分为3组,每组12只。对照组灌胃法给予生理盐水2ml/(kg·d),每周2次肌内注射生理盐水,每次0.5ml/kg;模型组同法给予生理盐水2ml/(kg·d),每周2次肌内注射地塞米松,每次2.5mg/kg;实验组同法给予辛伐他汀10mg/(kg·d),并同法给予等量地塞米松。8周后对各组动物作第三腰椎骨密度测量和组织形态显微结构观察以及左股骨骨组织形态计量学分析。结果实验组第三腰椎骨密度高于模型组(P<0.01),但与对照组比较差异无统计学意义(P>0.05)。实验组第三腰椎骨小梁形态结构与对照组相似,模型组呈骨质疏松症表现。实验组左股骨骨体积、类骨质表面、类骨质宽度和成骨表面均较模型组增高,骨重建时间缩短(P<0.01);实验组上述指标与对照组比较,差异无统计学意义(P>0.05);3组间骨吸收表面、矿化率、矿化时间差异无统计学意义(P>0.05)。结论辛伐他汀可有效预防糖皮质激素性骨质疏松。
Aim. To study the effect of simvastatin on preventing glucocorticoid-induced osteoporosis. Methods: A total of six-month 36 male SD rats were randomly divided into 3 groups,12 in each group. Group A was given normal saline 2 mL/( kg · d) through gastric tube,and normal saline at 0.5 mL/kg twice a week through intramuscular injection. Group B was given normal saline 2 mL/( kg · d) through gastric tube, and dexamethasone at 2.5 mg/kg twice a week through intramuscular injection. Group C were given simvastatin 10 mg/(kg · d) through gastric tube, and dexamethasone at 2.5 mg/ kg twice a week through intramuscular injection. After 8 weeks, the third lumbar vertebral bodies of all the rats were obtained and subjected to study bone mineral density and the structure of bone trabeculae, and the bone histomorphometry of left femur was analyzed. Results: The structure of bone trabeculae of Group C was similar to that of Group A, and Group B presented the feature of osteoporosis. Compared with those of Group B,the average of bone volume,osteoid surface,osteoid thickness ,and osteoblast surface of Group C were significantly higher, and the time for bone establishment was shorter (P 〈 0.01 ) , but the values of these norms mentioned above in Group C did not differ from those of Group A(P 〉 0.05 ). While no difference was observed in the bone absorption surface, mineralization rate and time for bone mineralization of three groups ( P 〉 0.05 ). Conclusion : Simvastatin can effectively prevent the osteoporosis induced by glucocorticoid.
