人脑胶质瘤组织中NF-κBp65蛋白及细胞周期检测

摘要 目的探讨人脑胶质瘤组织中NF-κBp65蛋白的表达及其与细胞周期的关系。方法应用免疫组织化学方法检测56例脑胶质瘤、8例正常脑组织标本中NF-κBp65蛋白的表达,用流式细胞仪分析胶质瘤细胞周期分布。结果胶质瘤组织中NF-κBp65蛋白的阳性率为67.9%(38/56),高于正常脑组织(0/8);Ⅲ、Ⅳ级胶质瘤组织中NF-κBp65蛋白表达阳性率高于Ⅰ、Ⅱ级组织(P<0.05),P65蛋白阳性率与瘤周水肿有关;而与患者年龄、性别及肿瘤大小等因素无关。随着NF-κBp65蛋白表达级别的增加,G0/G1细胞比例逐渐降低,S期细胞比例及细胞增殖指数PI逐渐增高。结论脑胶质瘤组织中NF-κBp65异常高表达,其表达与胶质瘤细胞增殖活性有关。 Aim: To explore the expression of NF-κBp65 protein in human glioma tissue and its clinical significance. MethOdS :The expression of NF-κBp65 protein in 56 cases of glioma tissue and 8 cases of normal brain tissue was detected using immunohistochemistry SABC method. Cell cycle was detected by flow cytometry. Results: The expression rate of NF-κBp65 in glioma tissue was 67.9% (38/56) ,higher than that in normal specimens (0/8) ;NF-κBp65 expression was wignificantly related to histopathological grade, brain edema grade, but not related to gender, age and the size of tumors. With the increase of the expression intensity of NF-gpB65, the G0/G1 decreased and S-phase fraction and proliferation index were iucreased significantly. Conclusion:The expression of NF-κBp65 is abnormally high in human glioma tissue, which is associated with proliferation activity of gliomas cell.
