曲古菌素A对视网膜母细胞瘤Hxo-rb44细胞凋亡及Sur-vivin、c-Myc表达的影响

摘要 目的探讨曲古菌素A(TSA)对人视网膜母细胞瘤(Rb)细胞株Hxo-rb44细胞凋亡和Survivin、c-Myc表达的影响。方法将人Rb细胞株Hxo-rb44进行培养传代,流式细胞仪检测不同浓度的TSA作用不同时间对Hxo-rb44细胞周期和凋亡的影响,用Westernblot法检测凋亡过程中Survivin和c-Myc蛋白表达的变化。结果TSA可明显诱导细胞发生G2期阻滞及细胞凋亡,而且与药物作用时间呈依赖关系(r=0.97,P=0.02);Westernblot法可以观察到Survivin和c-Myc的表达水平随着TSA作用时间的延长而下降(P均<0.01)。结论TSA可以有效诱导人Rb细胞株Hxo-rb44细胞周期阻滞和凋亡,其作用机制与抑制Survivin和c-Myc表达有关。 Aim : To investigate the effect of trichostatin A ( TSA ) on apoptosis and expression of Survivin and c-Myc of retinoblastoma cell Hxo-rb44. Methods:The retinoblastoma cell Hxo-rb44 was chosen in this experiment. The cell cycle arrest and apoptosis of Hxo-rb44 cell were assayed with flow cytometry. Expression of Survivin and c-Myc was detected by Western blot. Results: TSA obviously induced cell cycle arrest and apoptosis in time-dependent manner ( r = 0.97,P = 0.02). The expression of Survivin and c-Myc decreased with TSA incubation time (P 〈 0.01 ). Conclusion: TSA can effectively induce cell cycle arrest and apoptosis in Hxo-rb44 cell, which may associate with TSA suppressing the expression of survivin and c-Myc.
