蛋白激酶C在大鼠小肠缺血预处理中的作用 被引量:3

ISCHEMIC PRECONDITIONING PROTECTS INTESTINE AGAINST ISCHEMIA REPERFUSIN INJURY BY PROTEIN KINASE C ACTIVATION

摘要 目的:探讨蛋白激酶C(PKC)在大鼠小肠缺血预处理中的作用。方法:采用大鼠小肠原位灌流的缺血再灌注(I/R)模型,观察PKC抑制剂PolymyxinB和H_7对缺血预处理(PC)的肠道组织及血管床保护作用的影响。结果:PC明显减轻了I/R后肠粘膜病理损伤、MDA产生、IDH漏出(P<0.01),并减少了肠道微血管壁对伊文思兰的渗出(P<0.01)。PolymyxinH和H_7完全阻断了这种保护作用。结论:缺血预处理对I/R的小肠组织和血管床有保护作用,其机理涉及PKC的激活。 Objective:To invextigate the effect of PKC on ischemic preconditioning(IPC) of rat intes-tine.Methods:On the model of in situ intestinal ischemia/reperfusion (I/R) injury of rat. the effeets of PolvmvxinBand H-(PKC inhbitors ) on intestinaltissue and vasculature protection of IPC were ob-served. Results:IPC signifleantly.attenuated intestinal mueosal lesionand leakage of LDH.CD.low-ered inerease of MDA content in intestinal tissue and vaseular permeability to Evans blue dve.These protective effeets were completly.abolished by administration of PKC inhibitor .Polymysin B or H_7. Conclusion:IPC Droteets intestinal tissue and vasculature from 1/R injury and these protective effeets are mediated by PKCaetivation.
