摘要
采用超声技术提取中药刺五加中的乌索酸和齐墩果酸,并建立HPLC-PAD法测定了乌索酸和齐墩果酸的含量。色谱柱为Nova-Pak C18(3.9 mm×300 mm,4μm),流动相为甲醇∶水(体积比,88∶12),流速为0.8 ml/min,二极管阵列检测器检测,检测波长210 nm,柱温25℃。乌索酸线性范围为0.136~1.224μg,线性回归系数为0.999 5;齐墩果酸线性范围为0.052~0.468μg,线性回归系数为0.999 4。样品中乌索酸的平均回收率为98.1%,RSD为1.1%(n=5);齐墩果酸的平均回收率为97.5%,RSD为1.0%(n=5)。该方法操作简便,结果准确、可靠,可用于刺五加中乌索酸和齐墩果酸含量的测定分析。
Ursolic acid and oleanolic acid in Acantpanax senticosus (Rupr. et Maxim. )Harms were extracted with the ultrasonic technology. A determination method of ursolic acid and oleanolic acid was established with the high performance liquid chromatograph and photodiode array detector, The chromatographic column, Nova-Pak C18(3.9 mm × 300 mm,4 μm), methanol-water(88:12, V/V), mobile phase with 0.8 ml/min flow rate, the detected wave length(210 nm) and the column tempemture(25 ℃ ) were adopted. The calibration curve of ursolic acid was linear under the content of 0. 136 - 1.224 μg and the correlation coefficient were 0.999 5. The calibration curve of oleanolic acid was linear under the content of 0. 052 - 0.468 μg and the correlation coefficient were 0.999 4. The average recovery of ursolic acid was 98.1% and RSD was 1.1% ( n = 5), The average recovery of oleanolic acid was 97.5% and E^D was 1.0% (n = 5). The content determination of HPLC was accurate and reliable and can be used to determine the content of ursolic acid and oleanolic acid in Acantopanax senticosus Harms.
