芪丹通脉片对急性心肌缺血大鼠降钙素基因相关肽和内皮素的影响 被引量:10

Effect of Qidantongmai Tablets on calcitonin gene-related peptide and endothelin in rats with acute myocardial ischemia

摘要 目的研究芪丹通脉片(QDTMT)对急性心肌缺血大鼠的防治作用是否与增加血浆降钙素基因相关肽(CGRP)和减少内皮素(ET)有关.方法将60只SD雄性大鼠随机等分为6组空白组,模型组,单硝酸异山梨酯片治疗组,QDT-MT低、中、高剂量组.大鼠腹腔注射垂体后叶素致急性心肌缺血作为动物实验模型,观察心电图变化.用放射免疫法测定大鼠血浆CGRP,ET含量.结果CGRP,ET血浆含量在各组间有显著差异,其中模型组血浆CGRP,ET含量(分别为58.48±5.00和120.08±5.72)均高于空白组(分别为50.60±6.53和51.49±5.47)(P<0.05),QDTMT低、中、高剂量组CGRP血浆含量(分别为63.30±4.10,70.27±5.39,75.86±4.01)均高与模型组(P<0.05).QDTMT低、中、高剂量组ET血浆含量(分别为101.40±6.10,81.50±5.15,64.14±4.82)均低于模型组(P<0.05).结论QDTMT对缺血心肌的保护作用可能是通过促进CGRP的释放,减少ET的含量而实现的. AIM: To probe whether the protection of Qidantongmai Tablets (QDTMT) against myocardial ischemia in rats is related to the increasing content of plasma calcitonin gene-related peptide (CGRP) and decreasing content of plasma endothelin (ET). METHODS: Sixty SD rats were averagely divided into 6 groups randomly: control group, model group, isosorbide mononitrate treated group, QDTMT low dosage group( QDTMTL), QDTMT medium dosage group ( QDTMTM), and QDTMT high dosage group (QDTMTH). The experimental acute myocardial ischemia model was established by intraperitoneal injection of pituitrin to rats and the electrocardiogram was recorded. By means of radioimmunoassay, the contents of plasma CGRP and ET were detected. RESULTS: The contents of plasma CGRP and ET between groups had significant differences. The contents of plasma CGRP and ET in model group(58.48 ±5.00 and 120.08 ±5.72) were higher than those in the control group (50.60 ± 6.53 and 51.49 ± 5.47 ) ( P 〈 0.05 ). The content of plasma CGRP in each QDTMT group(63.30 ±4.10, 70.27 ±5.39, 75.86 ±4.01, from low to high dosage of QDTMT) was higher than that in the model group ( P 〈 0.05 ), while the content of plasma ET in each QDTMT group ( 101.40 ± 6.10, 81.50 ± 5.15, 64.14 ± 4.82, from low to high dosage of QDTMT)was lower than that in the model group ( P 〈 0.05 ). CONCLUSION : The protection of QDTMT against myocardial ischemia may be achieved by increasing the releases of calcitonin gene-related peptide and decreasing the content of endothelin.
