肺循环血流增加造成肺高压模型大鼠preproET-1mRNA和eNOSmRNA表达的改变 被引量:4

摘要 目的观察肺循环血流增加肺高压模型中前内皮素-1原mRNA(preproET-1mRNA)和内皮型一氧化氮合酶mRNA(eNOSmRNA)表达的变化。方法24只SD大鼠经颈部正中切口游离颈总动脉和颈静脉后随机分为3组(每组8只),对照组(C组):立即缝合切口、手术6周组(O6组)和手术10周组(O10组):经颈总动脉和颈静脉端-端吻合制作肺循环血流增加肺高压模型,O6组饲养6周,O10组饲养10周。至观察终点,测各组颈总动脉压、肺动脉平均压(mPAP),右心房血气,右心室肥厚指数,并取肺组织RT-PCR法做preproET-1mRNA和eNOSmRNA表达含量变化。结果动-静脉分流后复制了肺高压模型,引起mPAP、右心室肥厚指数升高;与C组相比,O6组和O10组preproET-1mRNA表达量逐渐增高(P值分别小于0.05和0.01),O6组和O10组差别无统计学意义;而eNOSmRNA表达量无明显变化。结论肺循环血流增加后内源性内皮素-1(ET-1)和一氧化氮(NO)改变是一个复杂过程,preproET-1mRNA表达增加与肺动脉血压增高和右心室肥厚并存,而preproET-1mRNA含量表达和eNOSmRNA含量表达相互之间无影响。 Purpose To investigate the different expression of preproET-1mRNA and eNOSmRNA in increased pulmonary blood flow in rats. Methods Twenty-four SD rats were randomly divided into 3 groups with 8 each. Control group(group C) ; with sham operation; rats in 06 group were performed left-to-right shunt operation which were obtained by carotid artery and jugular vein anastomosis and then lived for 6 weeks; rats in O10 group were performed the same operation and then were raised for 10 weeks. Then, under general anesthesia the systemic blood pressure (SAP), pulmonary arterial pressure (PAP), blood gas of right atrium and index of right ventricle hypertrophy(RVH) were measured. After that the tissue of pulmonary were obtained to measure preproET-1mRNA and eNOSmRNA by RT-PCR. Results The animal model of pulmonary hypertension was confirmed by increased PAP and index of RVH both in O6 and O10 group. Compared with group C, the expression of preproET-1mRNA in 06 and 050 group were significantly inereased(P〈0.05 and P〈0.01). The expressions of eNOSmRNA were not significantly different among the 3 groups. Conclusions With the increasing of PAP and RVH, the expressions of preproET-1mRNA were inereased at the same time in the pulmonary hypertension model of increased pulmonary blood flow. But the expressions of eNOSmRNA did not changed.
