摘要
目的探讨瘢痕内缺氧环境减轻的机制。方法采用免疫组化法检测烧伤后肉芽、不同时期瘢痕和正常皮肤中缺氧诱导因子1α(hypoxiainduciblefactor1α,HIF1α)、p53的表达,采用权重方法分别对各组瘢痕的HIF1α、p53表达结果进行量化,分析各自规律及相互间关系。结果随着瘢痕的成熟,HIF1α表达强度逐渐减弱,p53表达强度在1年内逐渐增强,相互间具有负相关性(P<0.01)。结论p53在瘢痕成熟过程中起重要作用,HIF1α可能通过协同p53诱导细胞凋亡,减少耗氧来减轻缺氧环境。
Objective To explore the mechanism of hypoxic environment lessening in the course of scar maturation. Methods The expression of hypoxia inducible factor 1α (HIF-1α) and p53 in granulation of burn wound, burn scars of different stages, and normal skin was detected with immunohistochemical staining. The expressions were quantified by the weight method. The relationship between HIF-1α and p53 was investigated. Results With the scar maturation, the expression of HIF-1α gradually decreased while the expression of p53 gradually increased within one year. There was a negative correlation between HIF-1α and p53( P 〈 0.01 ). Conclusion p53 plays an important role in scar maturation. Associated with p53, HIF-1α may induce apoptosis, which decreases oxygen consumption and lightens hypoxia in the scar maturation.
