干扰素-γ对人结膜下Tenon’s囊成纤维细胞表型转化的干预

摘要 目的探讨干扰素-γ(IFN-γ)对人结膜Tenon s囊成纤维细胞(HTCF)表型转化的干预作用。方法4例白内障手术中,取结膜下组织块培养成纤维细胞。用蛋白质印迹免疫检测技术和免疫细胞化学技术鉴定细胞表型。检测未经诱导及经转化生长因子-β1(TGF-β1)诱导(有或无IFN-γ预处理)后的HTCF表达α-平滑肌肌动蛋白(α-SMA)的情况。结果与未经诱导的HTCF相比较,10ng/mL的IFN-γ能显著抑制HTCF转化为肌成纤维细胞(MF)(P<0.01)。对于经过IFN-γ预处理后再用TGF-β1诱导的HTCF,IFN-γ仅能部分抑制其表达α-SMA。结论IFN-γ能抑制HTCF转化为MF;仅能部分干预TGF-β1诱导HTCF转化为MF的作用。 Objective To explore the effect of interferon-γ (IFN-γ)on phenotypic conjunctival capsular fibroblast(HTCF). Methods Cultured HTCF derived from 4 transition of human Tenon' s operated human cataracts was induced for 48 hours in absence or presence of IFN-γ and/or transforming growth factor-β1(TGF-β1 ). Then immunocytochemistry and Western blot technology were used to detect the α-smooth muscle actin(α-SMA)expression and identificate the cell phenotype. Results In contrast to normal HTCF, IFN-γ (10 ng/mL) inhibited the expression of α-SMA(P 〈0.01). It had little effect on inhibiting TGF-β1 to induce the expression of α-SMA of HTCF, which was pretreated by IFN-γ for 30 min. Conclusion IFN-γ may have negative effect with regard to controlling the myofibroblastic phenotype of HTCF itself.
