摘要
本文报导用气相色谱法测定人白蛋白制品中辛酸钠含量。样品用正庚酸作内标,经酸化、氯仿抽提、浓缩后,通过HP-INNOWax柱,以氢火焰检测器测定。本法色谱峰形好,平均回收率及变异系数分别为98.54%、1.39%。测定线性范围在107μg/0.25ml~720μg/0.25ml。最小检测限为7.410μg。实验操作简便,样品及溶剂用量少,可作为人白蛋白制品中辛酸钠含量的检测方法。
A quantitive gas chromatographic method for the determination of sodium caprylate in human serum albumin was reported The heptanoic acid was added in as internal standard,and the samples were treated with perchloric acid and extracted with chloroform.After being concentrated,the extracts were chromatographed on a 0.53mm HP-INNO Wax colum at 140℃ and determined with flame ionization detector for octanoic acid. There is a linear relationship of dose-response over the concentration range between 107 ug/0.25 ml and 720 ug/0.25 ml.The average recovery was 98.54%,and the average C.V. was 1.39%.The limit of detection was 7.410 ug.So this method can be adopted as the way for testing the contents of sodium caprylate in human serum albumin products.
